Each lens in a microscope has a numerical aperture, or NA, value. This has to do with the angles of light that enter and exit a lens. Its applications are beyond the scope of this lab, but numerical aperture does influence the resolution possible with a particular lens, and so the NA value for the lens is usually printed on each objective. In this video we explore what Numerical Aperture is in fibre optics, we look at its causes, what it does to a fibre optic link and we also look at what can b To achieve high magnification and resolution at the same time, a combination of objective and ocular lenses with numerical aperture or different light range angles is used.

Another way to look at this is by the concept of numerical aperture (NA), which is a measure of the maximum acceptance angle at which a lens will take light and still contain it within the lens. Figure \(\PageIndex{1b}\) shows a lens and an object at point P . For microscopic instruments, the diffraction-limited spatial resolution is proportional to the light wavelength, and to the numerical aperture of either the objective or the object illumination source, whichever is smaller. Resolution, Depth of Focus, and Depth of field • Resolution for coherent illumination: • R = k1λ/NA • R measured as minimum usage feature size in microns • k1 process factor; λ= exposure wavelength; NA numerical aperture • Resolution for partially coherent system • … Without getting too technical, the only way to get a Numerical Aperture greater than 1.0 is to use a material with a refractive index greater than 1.0. Oil (1.5) is such a material.

Numerical Aperture. NA depends on the objective construction, working distance, and the refraction coefficient n of the immersion medium: where α is the maximum light cone angle ( Figure 1 (a) ). Typically, the higher the objective magnification, the larger the NA (and therefore higher 2D resolution, r ).

The NA of a lens is defined as n(sin θ), where n is the refractive index of the medium between the specimen and the  Numerical aperture determines the resolving power of an objective, the higher the numerical aperture of the system, the better the resolution. Numerical Aperture and Image Resolution. The image formed by a perfect, aberration-free objective lens at the intermediate image plane of a microscope is a diffraction pattern produced by spherical waves exiting the rear aperture and converging on the focal point. 2017-06-07 Numerical Aperture and Image Resolution The image formed by a perfect, aberration-free objective lens at the intermediate image plane of a microscope is a diffraction pattern produced by spherical waves exiting the rear aperture and converging on the focal point. Numerical Aperture and Image Resolution - Java Tutorial The image formed by a perfect, aberration-free objective lens at the intermediate image plane of a microscope is a diffraction pattern produced by spherical waves exiting the rear aperture and converging on the focal point. Numerical aperture in electron microscopy has a very similar meaning to that in optical microscopy.

Without getting too technical, the only way to get a Numerical Aperture greater than 1.0 is to use a material with a refractive index greater than 1.0. Oil (1.5) is such a material. So to get the best resolution at 1000x (N.A. 1.25), a drop of this special oil is placed between the lens and the slide.

Numerical aperture: The numerical aperture of the lens typically increases with the magnification, starting at around 0.25 on a 10x lens, then 0.6 to 0.8 on a 20x lens, and 1.47 on an oil immersion lens. Magnification: The magnification we are referring to is only that of the objective lens and not the ocular lens.
